Background:
ACLY is the major enzyme responsible for the synthesis of cytoplasmic acetyl-CoA in many tissues. The product it catalyzes, acetyl-CoA, is used in several important biosynthetic pathways, including cholesterogenesis and lipogenesis.

Assay Principle:
The kit is based on a sandwich ELISA. The capture antibody is pre-coated on a 96-well plate. A biotin-conjugated antibody is then used as the detection antibody. Standards, test samples, and biotin-conjugated detection antibodies are then added to the wells and washed with buffer. HRP-streptavidin is added, and the unbound conjugate is washed away with washing solution. TMB substrate is used to visualize the HRP enzyme reaction. TMB produces a blue product catalyzed by HRP, which turns yellow upon addition of an acidic termination solution. The density of the yellow color is proportional to the amount of target sample captured in the plate. The concentration of the target can be calculated by reading the absorbance at 450nm with a microplate reader.
