Description: Malate dehydrogenase (MDH) activity assay kit (Colorimetric) is an enzyme assay kit for the rapid and sensitive detection of MDH. The detection limit is 0.5 U/L.
Background: Malate dehydrogenase (MDH) is an enzyme that reversibly catalyzes the oxidation of L-malate to oxaloacetate in the presence of NAD. There are two isoenzymes in eukaryotic cells: MDH1 and MDH2. MDH1 is found in the cytoplasm and plays a key role in the malate-aspartate shuttle that transports malate into the mitochondria. MDH2 is a mitochondrial enzyme that participates in the TCA cycle and reversibly converts L-malate to oxaloacetate.

Assay Principle: The tetrazolium salt MTT is reduced to reduced MTT in the NADH-coupled enzymatic reaction, which shows a maximum absorption peak at 565 nm. The increase in absorbance at 565 nm is proportional to the enzyme activity.
