Description: In situ histone H3-K27 tri-methylation assay kit is an epigenetic test that detects trimethylation of histone H3-K27 in situ under simple, reliable, and consistent conditions.
Background: Epigenetic activation or inactivation of genes is associated with a variety of diseases. A major mechanism of epigenetic inactivation of genes is the methylation of CpG islands in genomic DNA by DNA methyltransferases. Histone methyltransferases (HMTs) control or regulate DNA methylation through chromatin-dependent transcriptional repression or activation. They transfer 1-3 methyl groups from S-adenosyl-l-methionine to lysine and arginine residues of histones. G9a and polycomb group enzymes are HMTs that catalyze the methylation of histone H3 at lysine 27 (H3-K27) in mammalian cells. Trimethylation of H3-K27 is a facultative heterochromatin mark that promotes the recruitment of polycomb group proteins for gene silencing global H3-K27 trimethylation increased H3-K27 trimethylation has also been found to be associated with several pathological processes. Assay Principle: In this assay, adherent cells are cultured in conventional 96-well microtiter plates. After the experimental treatment, the cells are fixed and permeabilized. Trimethylated H3-K27 is then detected with an anti-trimethylated H3-K27 antibody. The proportion or amount of trimethylated H3-K27 is quantified by the intensity of the HRP-conjugated secondary antibody chromogenicity.
