Specification
Description:
Human medium chain specific acyl CoA dehydrogenase (ACADM) ELISA kit (0.5-10 ng/mL) is a ready-to-use ELISA kit for analyzing human ACADM levels with a detection range of 0.5-10 ng/mL and a sensitivity of 0.1 ng/mL.
Background:
ACADM is an enzyme that catalyzes the first step of mitochondrial fatty acid oxidation. It may serve as a target to regulate fatty acid metabolism for the treatment of obesity, cancer, and other related diseases.
Formula Weight:
47,020 Da
Assay Principle:
The kit uses a competitive enzyme immunoassay technique utilizing a polyclonal anti-ACADM antibody and an ACADM-HRP conjugate. The test samples and buffer are incubated in a pre-coated plate with the ACADM-HRP conjugate for one hour. At the end of the incubation period, the wells are decanted and washed five times. The wells are then incubated with substrate for the HRP enzyme. The product of the reaction of the enzyme with the substrate forms a blue colored complex. Finally, the reaction is stopped by the addition of a stop solution, which turns the solution yellow. The color intensity is measured spectrophotometrically at 450 nm on a microplate reader. The color intensity is inversely proportional to the concentration of ACADM because the ACADM in the sample competes with the ACADM-HRP conjugate for the binding site of the anti-ACADM antibody. Since there are a limited number of binding sites, as more sites are occupied by ACADM in the sample, fewer sites remain to bind the ACADM-HRP conjugate. A standard curve is plotted between color intensity (O.D.) and standard concentration. The concentration of ACADM in each sample is derived from this standard curve.
