Background:
AGRP plays a role in body weight homeostasis, is involved in the control of eating behavior through the central melanocortin system, and acts as an inverse agonist of MC3R and MC4R, thereby inhibiting their constitutive activity. Genetic variants in AGRP may be responsible for obesity.

Assay Principle:
The kit utilizes a sandwich-ELISA method. The microplates provided in the kit are pre-coated with an antibody specific to AGRP. Standards or samples are added to the appropriate microplate wells and bound to the specific antibody. The biotinylated detection antibody specific for AGRP and avidin-HRP conjugate are then added sequentially to each microplate well and incubated. The free components are washed away, and substrate solution is added to each well. Only wells containing AGRP, biotinylated detection antibody, and avidin-HRP conjugate will appear blue. Upon addition of sulfuric acid solution, the enzyme-substrate reaction terminates, and the color changes to yellow. Optical density (OD) is measured spectrophotometrically at 450 nm ± 2 nm. The OD value is proportional to the concentration of AGRP. The concentration of AGRP in the sample is calculated by comparing the OD value of the sample with the standard curve.
