ACAC (also known as ACC) is the enzyme that catalyzes the carboxylation of acetyl coenzyme A to produce malonyl coenzyme A, which is involved in fatty acid biosynthesis and β-oxidation. It is a potential target for the treatment of obesity and related diseases.

Assay Principle:
The kit is based on the sandwich ELISA technique. The anti-ACAC antibody is pre-coated on a 96-well plate. A biotin-conjugated anti-ACAC antibody is used as the detection antibody. Standards, assay samples, and biotin-conjugated detection antibodies are then added to the wells and washed with buffer. HRP-streptavidin is added, and the unbound conjugate is washed away with washing solution. TMB substrate is used to visualize the HRP enzyme reaction. TMB produces a blue product catalyzed by HRP, which turns yellow upon addition of an acidic termination solution. The density of the yellow color is proportional to the amount of ACAC sample captured in the plate. The concentration of ACAC can be calculated by reading the absorbance at 450 nm with a microplate reader.
