Description:
Histone H3-K9 methylation assay kit is an epigenetic test kit that uses a colorimetric assay to measure histone H3-K9 methylation and can be analyzed manually or in high throughput.
Background:
Histone methyltransferases (HMTs) control or regulate DNA methylation through chromatin-dependent transcriptional repression or activation. ESET, SUV39-h2, and others are HMTs that catalyze the methylation of histone H3 lysine 9 (H3-K9) in mammalian cells. H3-K9 methylation participates in the silencing of gene expression at nonchromatin sites and mediates heterochromatin formation through the formation of HP1 binding sites. Increased overall H3-K9 methylation has also been found to be associated with several pathological processes. Visual measurement of histone H3-K9 methylation is necessary.

Assay Principle:
In this assay, histones are stably spotted on bar wells. Methylated Histone H3-K9 is recognized by high-affinity antibodies. The proportion or amount of methylated H3-K9 can be quantified by an HRP-conjugated secondary antibody chromatography system and is proportional to the intensity of the chromatography.
