Description:
Histone H3-K27 tri-methylation assay kit is an epigenetic test that uses a colorimetric assay to measure histone H3-K27 tri-methylation, making the assay more flexible and efficient.
Background:
Histone methyltransferases (HMTs) control or regulate DNA methylation through chromatin-dependent transcriptional repression or activation. They transfer 1-3 methyl groups from S-adenosyl-l-methionine to lysine and arginine residues of histones. G9a and polycomb group enzymes are HMTs that catalyze the methylation of histone H3 at lysine 27 (H3-K27) in mammalian cells. H3-K27 trimethylation is a facultative heterochromatin marker that promotes recruitment of polycomb group proteins for gene silencing. It is also found that increased global H3-K27 trimethylation is associated with pathological processes. Measurement of histone H3-K27 trimethylation is essential.

Assay Principle:
In this assay, histones are stably spotted on the bar wells when the kit is used for detection. Trimethylated Histone H3-K27 is recognized by high-affinity antibodies. The proportion or amount of trimethylated histone H3-K27 can be quantified by the intensity of HRP-conjugated secondary antibody color development.
