Description: Histone demethylase (HDM) activity/inhibition assay kit (H3K4 specific) is an epigenetic test kit for H3K4-specific histone demethylase activity/inhibition measured by fluorescence assay using cell/tissue extracts and can be analyzed manually or in high throughput.
Background: Lysine histone methylation is one of the most powerful epigenetic marks and is critical for the regulation of a wide range of cellular processes. Methylation of histone H3 lysine 4 (H3K4) appears to be of particular significance because of its association with active regions of the genome. H3K4 methylation is considered irreversible until the discovery of a large number of histone demethylases suggested that demethylation events play a significant role. To date, at least two classes of H3K4-specific histone demethylases have been identified. Among them, JARIDs (e.g., RBP2, PLU-1, SMCX, and SMCY) catalyze the removal of methylation via a hydroxylation reaction that requires iron and α-ketoglutarate as cofactors, whereas LSD1 removes dimethylation and monomethylation from H3K4 via an amine oxidase reaction. H3K4-specific demethylases are found to be associated with pathological processes. Inhibition of this enzyme may lead to remethylation of H3K4 and silencing of H3K4-enriched active genes. Assays for histone demethylase activity/inhibition are necessary.

Assay Principle: The unique dimethylated histone H3K4 substrate is stably captured on the wells of the reagent strips during the assay. The active HDM binds to and demethylates the histone H3K4 substrate. The remaining unmethylated substrate is recognized by a high-affinity anti-methylated histone H3K4 antibody. The ratio or amount of unmethylated histone can be quantified by fluorescence, and ultimately, the HDM enzyme activity can be calculated.
