Description:
Di-methyl histone H3K4 quantification kit (Colorimetric method) is an epigenetic test kit that provides a tool to measure histone H3K4 global dimethylation. The assay conditions are simple, reliable, and consistent.
Background:
Histone methyltransferases (HMTs) control or regulate DNA methylation through chromatin-dependent transcriptional repression or activation. They transfer 1-3 methyl groups from S-adenosyl-l-methionine to lysine and arginine residues of histones. SET1, SMYD3, etc. are HMTs that catalyze histone H3 lysine 4 (H3K4) methylation in mammalian cells. H3K4 dimethylation is associated with transcriptionally permissive euchromatin regions of the genome and may be a global epigenetic mark in euchromatin and mediates activated transcription. It is also found that increased global H3K4 dimethylation is associated with pathological processes. Quantification of global H3K4 will aid in the development of HMT-targeted drugs and a better understanding of the epigenetic regulation of gene activation.

Assay Principle:
In this assay, dimethylated histone H3 at lysine 4 is captured in the wells of a reagent strip coated with an anti-dimethyl H3K4 antibody. The captured dimethylated histone H3K4 can be detected with a labeled detection antibody and then color-developed with a chromogenic reagent. The proportion of dimethylated H3K4 is proportional to the absorbance. The absolute amount of dimethylated Histone H3K4 can be quantified by comparison to a standard control.
