Description:
Cellular senescence assay kit (Flow cytometry) is a cellular senescence assay kit that provides an efficient method for measuring senescence-associated (SA) ß-gal lactosidase activity. Fluorescent substrate is added directly to senescent cells in 35 mm flat dishes. It can later be measured and analyzed by flow cytometry or epifluorescence microscopy.
Assay Principle:
The assay uses a fluorescent substrate to determine SA-ß-Gal activity. This compound is a membrane-permeable, non-fluorescent substrate for ß-galactosidase that emits green fluorescence upon hydrolysis of galactose residues and remains intracellular.
Background:
Cells stop growing during proliferation and develop a senescent phenotype. This limited lifespan correlates with the age of the organism and the expected lifespan of the species acquiring the cells; the older or shorter the lifespan, the less able the cells are to undergo population multiplication. Senescent cells are characterized by irreversible G1 growth arrest, including repression of genes that drive cell cycle progression and up-regulation of cell cycle repressors such as p16INK4a, p53, and its transcriptional target p21CIP1. They are resistant to mitogen-induced proliferation and show a characteristic enlarged, flattened morphology. Therefore, the study of positively regulated pathways of senescence and how cells bypass senescence is crucial for disease development.
