Overview

Obesity is a chronic metabolic disease caused by a combination of genetic and environmental factors. To further understand the pathogenesis of obesity and explore effective treatment methods, In Vivo Models and Cell Models are essential research objects. The dog is a Mammal and is similar to humans in anatomy, physiology, and pathological processes, which makes it an excellent Obesity Model. Protheragen, as a leader in the field of professional Anti-Obesity Preclinical Research, provides clients with personalized solutions with high efficiency and peace of mind.

Rely on Our Exceptional Ability to Build Unparalleled Dog Obesity Models

  • Dog Obesity Modeling

Diet-induced obesity model: Our researchers induce obesity by giving dogs a high-calorie, high-fat diet. The calorie and fat content of the feed needs to be precisely controlled according to the needs of the experiment. During the experiment, the dog's body weight, body fat content, and other relevant physiological indicators need to be monitored regularly to assess the degree of obesity.

Genetic obesity model: Certain breeds of dogs have a genetic predisposition to obesity, and these dogs are used as models for studying genetic obesity. Our researchers provide services to analyze the mechanism of action of obesity-related genes through an in-depth study of the genes of these dogs.

Drug injection-induced obesity model: Our researchers construct dog obesity models by plugging a wall with injections of monosodium glutamate (MSG) or aurothioglucose.

Bilateral ovary removal model: Our researchers establish an experimental animal model by performing bilateral ovarian removal in female dogs.

  • Professional Analysis

Our company provides comprehensive model-building and evaluation services. The metrics we test include, but are not limited to:

    • Body fat
    • Heart function and blood pressure
    • Feed intake
    • Inflammatory factors
    • Enzyme-linked immunosorbent assay(ELISA)
    • Western blot
    • Blood collection and analyses
    • Body weight
    • Colonic mucosal barrier function
    • Ultrasonography and oscillometry
    • Transmission electron microscope (TEM)
    • High-throughput sequencing

Workflow

The process of constructing the obese dog model is as follows: Firstly, our researchers select experimental dogs of appropriate breeds and ages and ensure that they have similar body weights, health conditions, and other basic conditions. Secondly, we induce obesity by feeding high-calorie and high-fat feeds, or by using interventions such as drug injections. During the modeling process, our researchers regularly monitor the body weights of the dogs, body fat rates, blood glucose, lipids, and other physiological indicators. Finally, our researchers perform relevant pathophysiology studies or drug testing depending on the different research needs of our clients.

Applications

  • The dog obesity model can be used in the drug development process to evaluate the therapeutic efficacy of novel con-obesity drugs for the treatment of obesity as well as possible side effects.
  • The dog obesity model allows for an in-depth study of the mechanisms of obesity, including genetic factors, environmental factors, and the interactions between them.
  • The dog obesity model can be used for rapid screening to identify obesity-related genes.

Advantages of Us

  • Our company carries out strict feeding management for experimental animals, including the control of feed formula, feeding environment, and the amount of exercise to ensure the accuracy and reliability of the experimental results.
  • The obese dog models offered by our company are not only suitable for the study of obesity itself but also the study of metabolic diseases such as diabetes and cardiovascular diseases.
  • We provide high-quality canine modeling services and comprehensive analytical services including, but not limited to, physiological indicators, pathological indicators, metabolite analysis, etc., according to the different research needs of our clients.

Results: In this study, researchers evaluated the distributional effects of ivermectin (IVM), moxidectin (MOX), and eprinomectin (EPR) using an obese canine model to see if obesity status affects the pharmacokinetic properties of these drugs. The results showed that obesity significantly affected the distribution and pharmacokinetic properties of the three drugs in dogs. In obese dogs, drug clearance was reduced, resulting in prolonged retention of the drugs in the body and increased blood concentrations. These changes may increase the risk of drug-related side effects in obese individuals.

Frequently Asked Questions

What are the advantages of dogs as lab animals?

The obese dog model has become an important tool for the study of obesity and its related diseases (e.g., diabetes mellitus, cardiovascular diseases) due to its high physiological similarity with humans such as food preference, psychological influence, and metabolic mechanisms.

What is the mechanism of drug injection-induced obesity modeling?

The mechanisms involved in drug injection-induced obesity models are multifaceted and include damage to hypothalamic neurons, disturbances in metabolic regulation, and alterations in appetite and feeding behavior. For example, injection of high doses of MSG damages the arcuate nucleus of the hypothalamus and its neighboring regions, leading to disturbances in feeding and energy metabolism and causing obesity.
